Les connaissances sur ce m\u00e9canisme ont des implications pour l&rsquo;am\u00e9lioration de la f\u00e9condation in vitro.<\/p>\n<p>Les \u00e9v\u00e9nements d\u00e9clench\u00e9s lorsque le sperme rencontre un ovule changent non seulement la vie des parents, mais sont \u00e9galement profond\u00e9ment fascinants d&rsquo;un point de vue scientifique.<\/p>\n<h2>La m\u00e8re de toutes les cellules<\/h2>\n<p>Apr\u00e8s la f\u00e9condation, l&rsquo;ADN de l&rsquo;ovule de la m\u00e8re et du spermatozo\u00efde du p\u00e8re constitue le sch\u00e9ma g\u00e9n\u00e9tique de l&#8217;embryon unicellulaire ou du zygote. L&rsquo;ADN paternel entrant contient des modifications qui facilitent une \u00abm\u00e9moire \u00e9pig\u00e9n\u00e9tique\u00bb de son \u00e9tat de sperme. Les prot\u00e9ines fournies par l&rsquo;\u0153uf f\u00e9cond\u00e9 agissent pour effacer en grande partie cette m\u00e9moire afin de g\u00e9n\u00e9rer un embryon totipotent qui peut donner naissance \u00e0 un tout nouvel individu. Les m\u00e9canismes sous-jacents \u00e0 la reprogrammation naturelle vers la totipotence sont remarquablement efficaces mais restent mal compris. \u00ab Pour mettre cela en perspective, la reprogrammation vers la pluripotence induite en culture cellulaire prend plusieurs jours \u00e0 plusieurs semaines alors que la reprogrammation vers la totipotence chez les zygotes se produit en moins de 24 h \u00bb, explique Kiku\u00eb Tachibana-Konwalski, qui consacre les recherches de son laboratoire \u00e0 la compr\u00e9hension des secrets mol\u00e9culaires de ovules et zygotes.<\/p>\n<h2>Nouvelle vie, nouvelle \u00e9pig\u00e9n\u00e9tique<\/h2>\n<p>Rapportant des recherches dans la revue scientifique Cell, des scientifiques de l&rsquo;Institut de biotechnologie mol\u00e9culaire (IMBA) bas\u00e9s \u00e0 Vienne ont d\u00e9couvert que non seulement les ovules f\u00e9cond\u00e9s d\u00e9clenchent une reprogrammation \u00e9pig\u00e9n\u00e9tique de l&rsquo;ADN du sperme, mais que ce processus est \u00e9troitement surveill\u00e9 pour pr\u00e9server l&rsquo;int\u00e9grit\u00e9 g\u00e9nomique.<\/p>\n<p>\u00ab\u00a0Lorsque le sperme p\u00e9n\u00e8tre dans l&rsquo;ovule, la chromatine m\u00e2le dens\u00e9ment compact\u00e9e doit \u00eatre enti\u00e8rement\u00a0\u00bb d\u00e9ball\u00e9e \u00ab\u00a0et restructur\u00e9e autour d&rsquo;\u00e9chafaudages prot\u00e9iques appel\u00e9s histones\u00a0\u00bb, a expliqu\u00e9 Sabrina Ladst\u00e4tter, premi\u00e8re auteure de l&rsquo;\u00e9tude. \u00ab\u00a0En utilisant des \u0153ufs de souris f\u00e9cond\u00e9s, nous avons montr\u00e9 que l&rsquo;ovule d\u00e9clenche activement la d\u00e9m\u00e9thylation de l&rsquo;ADN paternel &#8211; en d&rsquo;autres termes, il initie une reprogrammation \u00e9pig\u00e9n\u00e9tique en supprimant toute m\u00e9moire \u00e9pig\u00e9n\u00e9tique ant\u00e9rieure transmise par le p\u00e8re. Cela permet au zygote de recommencer \u00e0 z\u00e9ro et de cr\u00e9er sa propre m\u00e9moire \u00e9pig\u00e9n\u00e9tique et son histoire de vie. Ce processus n&rsquo;est pas sans risques : la d\u00e9m\u00e9thylation peut provoquer des l\u00e9sions de l&rsquo;ADN qui peuvent \u00eatre mortelles pour le nouvel organisme. On sait que ces l\u00e9sions peuvent conduire \u00e0 la fragmentation des chromosomes, \u00e0 la perte d&#8217;embryons ou \u00e0 l&rsquo;infertilit\u00e9.<\/p>\n<h2>Un point de contr\u00f4le mol\u00e9culaire<\/h2>\n<p>Les chercheurs ont identifi\u00e9 un m\u00e9canisme de surveillance qui non seulement surveille les l\u00e9sions de l&rsquo;ADN caus\u00e9es par la reprogrammation \u00e9pig\u00e9n\u00e9tique, mais r\u00e9pare \u00e9galement les dommages. Ils ont r\u00e9v\u00e9l\u00e9 que les l\u00e9sions de l&rsquo;ADN paternel caus\u00e9es par la d\u00e9m\u00e9thylation activent un \u00abpoint de contr\u00f4le\u00bb zygotique qui emp\u00eache la division cellulaire jusqu&rsquo;\u00e0 ce que ces l\u00e9sions soient r\u00e9par\u00e9es. Ce m\u00e9canisme garantit donc que la reprogrammation est termin\u00e9e dans un cycle cellulaire et prot\u00e8ge l&rsquo;int\u00e9grit\u00e9 g\u00e9nomique au stade embryonnaire monocellulaire volatil. Fait int\u00e9ressant, ils ont \u00e9galement constat\u00e9 que les conditions dans lesquelles les embryons sont cultiv\u00e9s affectent la rigueur de la r\u00e9ponse au point de
